Within the Leibniz Graduate School on Aging (LGSA) in Jena the Neri Research Group invites applications for a PhD student position (TV-L E13/2).
The main goal of the present project is to characterize epigenetic changes that occur in calorie-restricted intestine and to delineate their role in changing stress responses during aging and inflammation. After genome-wide DNA methylation profiling conducted on intestinal stem cells (ISCs) and crypts, identified DNA methylation changes will be functionally characterized by using in vitro models (intestinal organoids) to assess the impact of these epigenetic alterations on the stem cell self-renewal, differentiation and response to inflammatory factors and chemical compounds. Targeting of locus-specific DNA methylation levels will be achieved by using a novel CRISPR-based editing approach. Epigenetically modified organoids will be phenotypically analysed as well as characterised at transcriptional level by using well established methods like RNA-sequencing.
The FLI is excellently equipped with state-of-the-art facilities like next-generation DNA sequencing (NGS) technology, proteomics, functional genomics, imaging and more.
